talk to the wife first, i really doubt shes blind to it, shes just helping her sis out. maybe she is getting fed up with it too, or at least can see how its hurting more than helping. after you get on a common ground with her, then both go to the sis and get the subject out in the open. either way, its not going to be all flowers and hugs, the sis is gonna feel dejected, mad and probably dissed.

You can't say anything to the twin and keep the peace. Your wife needs to do it. This will not end well if you confront them.

You should talk to your wife first. If she feels the same way, then you can talk to them together. If she doesn't care, then you are on your own. If you don't give a shit if they like you or not, tell them to get out.
 
Step up and be a man if you don't you will always feel resentment. Talk to your wife and tell her how it going to be and if she don't like that just to bad for her. If you don't do this I'm pretty sure you will be back to bitch some more.

It is early in your marriage, but you may need to get a few things ironed out.
1) Always be your spouse's ally. always. If you disagree do not do it in front of others.
2) Don't assume anything. discuss stuff ad nauseum if you have to but it will make for good times in the future.
3) Don't go to bed with anything bad between you. Settle all that you can, sacrifice sleep, talk it out.

Looking at those three rules, you are basically screwed this time around.
Anything you start on your own will probably violate one or more of those tenets. Unless you can get your wife to agree to put up a unified front to her sister's behavior while she is there, you will have to wait and discuss it after she leaves.
