My all time favorite line:

Grand Theft Auto: San Andreas had some truly hilarious dialogue. I've probably never laughed so much while playing a game. Escape from Monkey Island was also a pretty funny title (didn't finish it though).

I would think when the concept of funniest is questioned that games like Conker's Bad Fur Day and Bard's Tale would be mentioned more. Or are we only mentioning games that were funny and not intending to be?

HK47 was awesome!
The first Max Payne was great for listening to the enemies conversations as you came up behind them.
Same with NOLF 1&2
I also loved Full Throttle, who can't help but laugh at post-apocolyptic country music.
