I normally road bike when I don't have much time (if I have time I usually load up the mountain bike and hit trails), so I just go up Old Tunnel from my place near Rockridge BART and then back down. It's a nice quick workout. Or if I have a bit more time, I'll go up Old Tunnel, ride along Skyline, down Redwood heading East, then back up to Skyline on Pinehurst and home. If I really have time I head out to the top of Mt. Diablo and back, and my favorite 100 mile Bay Area route is up Mt. Hamilton, down the back side, North through Livermore, then return past the Calaveras Resevoir.
There are a billion great road rides around here in the East Bay. The Oakland Yellowjackets have a good bunch of route sheets on their web site.
Your profile says you're in SF. Plenty of great riding to be had by going across the GGB into Marin.
