Red Faction: Anytime you can strap a remote control explosive to the back of someone's back. Only to have them notice and flayle around, arms waving, screaming, "get it off, get it off" Deserves a laugh from me. . .

Black and White by far the game I got the most laughs. I laughed so hard when my creatured proceded to poo all over the village, ruining monuments, and buildings. Could you image what the villages were thinking? OMG the sky is raining poo, what did we do to deserve this. Then I just about died when my creature was stupid enough to actually try eating said poo.
 
I had a few laughs with the Gabriel Knight games.

The most I laughed was while playing the Tex Murphy games. Under a Killing Moon (I was laughing starting with the intro movie with James Earl Jones as God) & the Pandora Directive. I missed Overseer and lost under a killing moon. I keep hoping that I'll stumble across a copy of each of them some day.
 
Definitely the funniest moment in my gaming experience is in N.O.L.F. 2, when the big Irish guy (spoiler warning) grabs the ominous, cloaked French assassin and it turns out that he's a midget on a unicycle. I don't even remember much of the following tricycle chase because I was laughing so hard.

Although sheer hysterical rolling on the floor laughter can come from some of the most unlikely places - I remember playing Nagano Winter Olympics for the N64 with some friends and we'd do the long jumps without learning how to play and we'd usually royally screw it up with incredibly amusing results. Then there was the first time playing GTA3 and going on firetruck or bus rampages was just so ridiculous (considering no game up to that point really ever let you do something so awful) we couldn't stop laughing.

I remember renting that Olympics game. I don't know what it was about it but I remeber laughing so hard.

ff the top of my head, I would say The Longest Journey.
 
Giants! I wish I could find that game to buy, my friend had it and the cut scenes were freaking classic. Armed & Dangerous was good too.

Leisure Suit Larry was awesome, I was like 13 and somehow got my dad to buy it for me, all the sequels were funny too.

daam I need to hop on Ebay and look for Giants I gotta buy it.

 
My favorite:
Star Control II - The interactions with the alien races were comedy gold.

I'll also have to agree with the Sam & Max Hit The Road suggestion.

are you serious? ... Giants: Citizen Kabuto ... i still have it .. funny as hell and - right now - i am playing Armed and Dangerous, alternating with RE-4

but i never finished Giants and won't ... i don't like playing games without "anytime save"🙁

PM me if you want to trade ... only possible isssue is that i don't see the manual ... just the original CD jewel case with two clean CDs ... so am unsure if it needs a CD key or the manual. ... or if i can find it

Agreed.

Shadow warrior was good too (as others mentioned), but NOTHING came close to duke when it came to laugh-fests whilst gaming.

first game that ever made me laugh outloud, like where that guy runs his skateboard over the mayors wife's dog and ***spllaatt**

monkey island 3 was very witty and psychonauts had its moments, like the kid with the squirrels watching him all the time
