They have paid tours that travel by bus. Varying costs and durations. Those are really good for getting an overview of the City and what it has to offer.
Mt Vernon is nearby and is a pretty good way to kill a chunk of time.
It's also is bitch to drive in DC, so you probably will be using Metro. wmata.com
Reagan Nation is the most "accessible" Airport.
Dulles and BWI are in the burbs and not close to DC at all. If you don't really care about costs and are using a rental car, the difference isn't as big but it is some extra travel time.
The Virginia side has better views of the falls but you can easily visit both sides if you have a car. The Maryland side has a rocky portion of the Billy Goat Trail if you are interested in hiking. Avoid both parkways during rush hour. They don't have a shoulder so a simple accident turns into a major backup.

If you make it out to the end of the runway at Dulles, the National Air and Space Museum Annex is totally worth a stop. They have an SR71 Blackbird and a Space Shuttle on display there....among quite a few other planes.
