No. Not one bit. People who CAN play guitar actually have more trouble playing, from what I've seen. The only benefit it does give is that it may help loosen up your fingers and teach them to move in somewhat awkward and uncomfortable ways.

I have Guitar Hero III but I still think Danzig - Mother from II is my favorite. The inclusion of so many master tracks in III is awesome though. I'd say Social Distortion - Story of My Life is my second favorite song.
