Sure, as long as both are NTFS. I don't know why Windows has that limitation, but it does. If you reinstall Windows you'll likely have to remount them all, but that's it.

If you reinstall Windows the settings are lost and they will get mounted as individual drives until you remount them in the folder. This is very useful for database servers that will have a lot of LUNs attached to them.
