Using Airport Express and Ethernet Connection

I have a Macbook Pro that has a wired and wireless connection. I can connect to two different networks with the two connections. Is there anyway I can use both connections at the same time.
 
I did this just the other night. I had my MacBook hooked up over ethernet to my desktop as I copied files back and forth. At the same time, i used the wireless on my laptop to surf the web and send an email.

If both connections lead to the same router however, OS X defaults to the fastest connection available. So, if I have my laptop wired and wireless-ed into my router, it will use the wired connection. There is then a brief hiccup while it switches connections if I unplug the wired.
 
I think my confusion stems from your wording, the Airport Express is a router. The Airport Card is what is in your Mac.

So you want to connect to two networks simultaneously, via your Airport Card & your Ethernet connection?

It is generally easiest to have the wireless connected first, then you connect the wired. So you connect to Router A wirelessly, then plug into Router B.

It will do it automatically. If you unplug the ethernet the wireless (if left on) will become your new internet connection. It should not causing any problems with transfers or downloads. The same is true in the inverse, with one exception, any transfers that you were doing over wireless will continue on wireless.
