Canned air.

Go outside ( i know, the horror  ), & blow the dust out on your balcony or backyard, etc.

I can't even count the number of times I have used a vacuum to clean a pc running or not. Not a single problem. It has to be in the hundreds. For the real stubborn dust a blast from the compressor with the vacuum behind it to catch it.

i do this in conjunction with a 1 foot blower fan on full blast. it blows most of the loose dust out in seconds leaving me to find the more hard on stuff with the compressed air canister, and it keeps the airflow going so that the dust doesnt just settle back down before i go inside again

ed: on another note, if you are taking the CompTIA A+ hardware certification test, and you were to answer the question asking the best way to clean a computer is with a vacuum cleaner, you would be marked down. just some food for thought.
