I assume you mean when the motherboard is turned off. If it is turned off, I don't see why not. Just unscrew the screws and remove the 3 pin power connector and put your own fan on and screw it in and put the new fan's power connector in.
Also, do be careful not to exert much downward force on the fan screws. They're at the corners of the heatsink, which applies uneven leverage to the corners of your CPU core. I've done the swap with my GC-68 without removing it, but I greased the screws so they turn with less torque, allowing for less downward pressure by my screwdriver to prevent slippage.
greasing the screws on that heatsink is a good idea, i'm going to try that. also, a copper shim for your cpu is also a cheap safety device an a very good idea
I would be very careful. Those screws are in there pretty good, and make the the heatsink doesn't try to turn on you when you're doing it. I've never tried to change the fan while it was on, buy my friend has and he told me the heatsink wanted to rotate on him while he was doing it.
