Only if you run one GPU, and then really only if the GPU is single slot width, because if your GPU is double width, the X-Fi is going to be in the middle slot and right up against the GPU's cooling, blocking its airflow. If you still had one GPU and moved the X-Fi to the 3rd slot, you'd cut the PCIe lanes for the GPU down to x8 from x16.
