I submitted my resume to a company I interned at for two summers. I also emailed the HR lady letting her know that I'm very interested in the job. She emailed me back saying she's not HR for that area anymore and that insert new name is the new HR lady. I never met her. Should I email the new HR lady expressing interest in the job or wait? I don't think a cover letter/email will be necessary as the job posting only accepts resumes. Maybe just a little introduction?

Cover letter is always necessary.

And yes, you should contact HR, along with the people you worked for previously.
 
Is it really that necessary for this situation? Any person who never worked here has no contacts so if they apply for a job all they get to do is submit their resume.

yes, this is why you have an advantage by getting to contact them
 
The best idea would be to send your resume to your old supervisor(s), and have them submit the application for you. They can put in a good word and would be more familiar with the new HR lady. You are not 'any person who never worked' there. You are the person who worked there for 2 internships. You have an advantage, use it, f*** the other people, they'd do it too if they had the chance.
 
Yes, contact her, contact your old supervisor to let them know you're looking. Imp is right, anybody else that has a connection to get the job they want would use it.
 
I'd second submitting it to the supervisor. The HR person is probably getting flooded by hundreds of resumes and cover letters right now, and yours will end up in the circular file if it doesn't get some additional help by someone within the company.
