PC with Windows 7 does not boot

I just installed and updated the Microsoft Office 2010 software. The system told me that it needed to reboot. Then it was stuck in the closing mode for 5 minutes. I switched the system off and now it does not reboot.

Where should I start looking?

I tried that already but the PC does not read the DVD.

The issue started when I was turning the computer off. It remained at the stage "shutting down" for 10 minutes.

There is power. The light of the DVD reader goes on. There is a hum from the fan, I believe.
i do not know about the hard drive.

I will power the computer with the case open and see if the fan turns and if i can feel or hear the hard drive.

Could it be a board failure? What else could it be?

JUst like Corkyg........but you already did that. take a closer look to your hard drive connection unplug.look for loose wires.if not the drive is defective.if you can try it on another pc.
 
Thank you Corkyg and Denis280 for your help. I found a great tool in the Asus web site entitled "Guidelines to solve a PC start up problem" that guided me to determine that the RAM memory was bad.
