I am setting up a P4 2.53 system and bought a new HSF for it. The HSF has some thermal tape on it. Should I, in addition to the tape, add some Acrylic Silver to the CPU?

Of course...🙂 Just didnt know if it should have been only one or the other. Or even if it would interfere with the termal tape.

You have to remove the tape before you apply the thermal compound. Sometimes its easy to remove but otherwise scrape it off with a hard suface sponge or anything that works and then clean it with some isopropyl alcohol or make sure the surface is clean, and then apply the thermal compound. Do not use both, either or (compound is better).

Silly, I've tried it both ways. It works just fine with Artic Silver on top of the tape. That's the way my daughter's is running now, at 31-32C idle, with a slightly overclocked Palamino. You will get slightly better temps if you remove the tape, though, since the thermal compound will be in direct contact with the hsf.
