Just use it. All "conditioning" may do is correctly calibrate a sensor, and/or make the voltage curve match what a sensor expects.
So, if it has sat on a shelf for awhile, it may not read the battery's level correctly for a few charges, since the level will likely be measured by the current voltage.
Several full discharges in a row are not worth that risk, IMO, when regular use should do the same amount of good. One full discharge, if the device calibrates itself to the battery's behavior, might be useful, since it could, with info from the old battery, be telling you your battery is much lower than it is, or worse, tell you it's fine when it's dangerously low. 3-4 cycles, though, is just being cruel to the battery. If you always top off your battery well before it can discharge low, don't even worry about it for one cycle.
