Invalid System Disk problem

I've got a pc that is rebooting overnight and displaying this error:

Invalid system disk, replace the disk and then press any key.

It's not doing it all the time...just periodically. It has done it the last to nights. I've run SpeedFan on it and I'm not getting any type of SMART errors. I did have Diskeeper 10.x set to auto defrag my hd's at night and kind of wondered if that might be playing some kind of a part in it. I have turned that off and it is still doing it.

Suggestions?

The hd has errors on it - what kind we dont know - normally a chkdsk will fix the problem but the corrupted files are basically zapped - so it is a good idea to backup vital files before doing this. It also could be a failing hd, so a thorough diagnostic run using the manufacturer's utilities would be a good idea - Seatools/Datalifegaurd/Maxblast etc.
