How to format a new makeshift external HD?

I jumped on a $90 400gb seagate deal with the intention of making it into a portable external drive, and bought an enclosure (not at that price or from compusa, of course) for it.

My question is: how do I format it? Can I just stick the drive into the enclosure, turn it on, and format it through explorer? Or is there a better way?

In WinXP, if the drive isn't already formatted, you can access it through the Computer Management applet and do a partition and format there.
