Probably about 30-45 minutes. Most of that time spent fumbling with the little fvcking usb, led, firewire, etc headers. Annoying little bastards :|

Realistically, it often takes over an hour to assemble a full computer from scratch and install Windows on it. But over two hours and you're doing something wrong.

Heres a simple breakdown.
5 minutes to figure out how to get the new case open. 1 if you had experience with the case before.
10 minutes to open the motherboard box and install the board.
5 minutes for CPU and fan install.
5-10 minutes to option the board(if board has jumpers) includes skimming thru manual.
5-10 minutes to install and option drives.
2 minutes to install memory and AGP and PCI cards.
10 minutes to connect and double check all connections.
4 minutes to go thru the BIOS.
1 minute to put case
This is just an estimate.

def. takes an hour to do the hardware part if your doing a quality job. I could prob. throw some ish together in like 20-30 minutes though and get it to boot. But if I take time to reroute cables, spread some nice thermal paste on the HS, etc. it takes me a good 1hr-2hr, provided I don't get stuck along the way with some crazy problem.
