Has an employer ever asked for your college transcript?

yup. in my first real job.

he said it was strictly procedure.

it was to ensure that i actually graduated from the course I graduated from.
 
I was asked verbally during screening interview what my GPA was. Never had to show a transcript, but did have to show proof to the HR that I graduated from university.
 
If you're in a technical profession (one of the natural sciences or engineering), you'll almost certainly get asked for it in your first few years out of college, and possibly afterwards too.

In non-technical professions, its still possible (especially for the higher-paying jobs), but not nearly as likely.

Only for an internship. After that I had one year of real-world, post-grad experience and the only crap that mattered to get my next job was my resume and what my managers said about me.

I did have to give them a copy of my college diploma when I was hired as a software developer. I guess it goes in my personnel folder as proof that I do in fact have a degree.
