Computer Slowdown

In the last day or two, my computer has developed a very noticeable slowdown doing just about anything. Click a link or icon and it takes a second or two for a response to occur. I done the obvious, like defrag, run CHKDSK, etc. but so far, nothing helps. What diagnostics that I have done so far show no problems, but I'm continuing to look. Any ideas that might speed this procedure up?
