Will both ends seal off as the fitting disconnects?

I am building a case that is in 2 parts, and I want to be able to disconnect the hoses at a certain point and remove the top part for servicing without having to drain the loop if possible.
 
No, they don't work the way I think you think they work. They don't "click" together, the hose goes through the nut then over the barb and the nut screws down over the barb. You may be able to do something like what you want though with some close off valves in the area you are wanting to seperate. You would still need to rebleed the loop though afterwards.
 
I would suggest looking at Koolance Quick Disconnects for what you are wanting.

Actually, the Koolance quick disconnects at Performance PC's were exactly what I was talking about. I just wasn't sure I could disconnect while the loop is full and have both ends cap off automatically.
