Is this doable? I imagine it would be. Can I just clone with Macrium (willing to try some other software) and have the new clone keep the drive letter I previously had so that all shortcuts to that drive still work?
This is just an additional internal storage drive, not the C drive. There are a few games and programs installed to it and it's the drive I host a media server for video and audio. I just want to make sure anything linked to that letter and location will still do so after the new hdd is cloned.

Fantastic, thanks. I'd hate to have my media renderers freak out because they somehow detect the hdd change and can't access the music or video.

Windows dynamically assigns drive letters during boot up and assigns it mostly based on what drives it finds and in what order.

You don't even need to clone it with special software, just format to the filesystem you want (if it isn't already) in windows, copy the files over (standard windows' file copy), then change the drive letters in Disk Management.
