No, you don't have to even though the instructions may direct you to do it.

Just hold the backplate in place and install the cooler. If you do this, you will have to remove your motherboard every time you want to remove the cooler to redo the thermal paste or anything else.
You will have to remove all the cards and disconnect all the motherboard cables and remove the screws ..... That is too much trouble

Alternatively, you can follow the instructions in the link I provided on the previous page and you will not have to remove the motherboard.
 
OK, I'm prepared to remove everything if changing paste, that's cool. I love taking stuff to bits! I'll be using my fave paste, MX-2. It's good for 8 years, apparently. Not that I'd leave it that long or stay with one processor until 2016, but whatever.
