Could someone clarify what the differences are between the three versions of the P5K? Or rather, what are the differences that actually matter? Do they justify the price differential?

Goto the asus site and read about them. You can do a compare of models I believe. P5K Deluxe is the best one... I think it has more sata ports... probably more overclocking options... not entirely sure

I think the difference, like the P5B-E vs P5B-Deluxe is the Deluxe has solid caps, 8 layer PCB (more stability?), more SATA (I think, can't remember) and a second PCI-e 8x slot or something like that.

The P5K and P5K-E both use solid caps. The P5K-E also features the same 8-phase power design that the deluxe
features. The only glaring difference I notice is the lack of a second gigabit NIC on the P5K-E.

EDIT:
In addition to the heatpipe difference as noted in the next post, the MOSFETs on the deluxe
also sport small heatsinks.

P5K Deluxe vs. P5K-E

Features:
2 Gigabit LAN ports (Marvell PCIe+Realtek PCI) vs 1 Gigabit LAN port (Marvell PCIe)
Heatpipes: P5K Deluxe | P5K-E /Slight difference

Accessories:
4 SATA cables vs. 2 SATA cables
1 x 2-port USB2.0 / 1-port IEEE1394 module vs. N/A
'Image Editing Software' vs N/A

That's all the differences I'm seeing between these two. The vanilla P5K is a little less featured. You can check the specs on the site.

Why wait any more if you have a P4? Buy a P5K now and get an x38 later if you really want one.

The problem with switching out to a X38 later is that motherboard swaps are a PITA.

Nah. Regardless, a P5K would be fine for now. I'm willing to bet the cost of X38 and DDR3 will scare a lot of people into staying with P35 anyway.
