Access Denied for Admin

I'm having an issue with one folder on my shared drive. Everyone in the domain is getting access denied. I tried to access it through domain admin account, Administrator account, and through local Administrator account. All get access denied. There are two users that say they accessed it this morning with no issues. I had to restore from backup to a different folder. Permission on there are normal. Everyone has change, and domain Admins have full. The only thing I didn't do yet is reboot the server which I would not like to do during production hours, but if I must, I must.

Anyone have any ideas before I kick everyone out of their work?

Have you tried taking ownership of the folder with the domain admin account and then giving yourself full control permissions?
 
You said you gave everyone permission to Change the contents of the folder under the Sharing tab but did you make sure to giver them access locally under the Security tab? I know this is something simple but I have overlooked it many times before when sharing folders with NT/2000/XP.
 
Someone deleted the folder!!! But for some reason the folder got stuck. After I rebooted the server the directory was gone. Sorry for the confusion. But thanks for the effort!
