Much has been made about Fable III with speculation that it will allow you to live the life of a king, support Project Natal, and even "piss you off" according to game designer Peter Molyneux.
What we have learned so far from Microsoft's X10 press event is that Fable III starts out with you playing as the 18-year old son of the character from Fable II and you have to fight to get your throne back from an usurper. But the game does not end there as you'll have to undertake the burden of ruling your kindgom once you get it back. And, to paraphrase Peter Molyneux, we often see rulers change visually their first year or two in power.
Other changes in Fable III includes a new morphing system. In Fable II, your character would change as you progressed through the game depending on your actions. Do evil things and you look more sinister. Do nice things and you'll look the part. Lionhead has thrown out that system and replaced it with a more extreme system that affects not just your character but the weapons he wields too. The weapons will change and adapt to your alignment. Evil swords will glow and drip with blood, for example.
Another big change brought to Fable III is the ability to touch other people. You'll be able to hold hands with your girlfriend or even pull a vagabond to a factory to sell into slavery.
