I have to say ReadyBoost has made a good amount of difference in my current system. Currently I am only using 2GB of DDR2-800 RAM and was going to add another 2GB stick later. I didn't want to put the extra money out right now so I threw in a 4GB flash drive for ReadyBoost and damn let me say it has made a difference. My system is a bit faster and more responsive and Star Trek Online does run a bit better. I also don't hear my hard drive grinding as much as it was either.....
