I installed Win10 1607 64-bit recently (clean install on an SSD), and she's had this problem a few times now. It's resolvable by rebooting (or - I think - signing out and back in again), but that sucks as a solution. As it's a desktop, it spends most of the time in sleep mode rather than being shut down, and my wife likes to leave stuff running.
I'm aware that there was an issue with Win10 RTM whereby nothing on the taskbar worked, but I can click on the Windows Defender icon, and Classic Start Menu is working fine. Any ideas?
I'm just running dism /restorehealth on it at the moment.
Admittedly I'd place a fair bit that the so-very-interesting single notification is going to be that Defender did a scan and didn't find anything, but it would be nice if the Action Center actually worked.
