I thought second is correct. But when I look up Quintet in dictionary, it says "Composition for five voices or five instruments". I thought it's "composition of something"?

They mean a composition intended for five voices to sing it. They refer to instruments in the same way.

I thought it could be either. The music written for 5 voices or instruments OR the group of them together. Maybe the groups of them together can be called that because the music is written for them.

I wrote this composition for five voices as a way to express a range of techinques, but the amazing range of the Diva of Plavalaguna in the film The Fifth Element was achieved through a composition of five voices.
