what the most efficient way to cool down a room?

Need more information. Is the room open to other rooms? Is there AC running? Is there a window open? Is it cooler or hotter outside than inside?

f you have two windows open and one fan, is it better to put the fan by one window with the fan blowing out or in???

Does it make a difference?

If one fan, you would want it to blow out. This will create a vacuum and draw the cooler air into the building.
 
Fan pointed out the door of the room.
The air will pull cool air from outside, door closed is useless.
