ASRock 890GX Extereme4 AM3 motherboard.

I have put a couple of cards in different slots, one video card and one sound card, one at a time breadboarded and computer will not post. Take out the card, no problem.

PCI slot lights up the sound card, but computer will not post.

Everything works without a card in the PCI slot. Running Win 7 Ultimate 64 bit.

Any guesses?

It's defective, return it. Thats assuming the cards work in another PC.

Don't have another PC to try them in, however I think you are correct.
