You want to express these primary things:
- Happiness that your best friend has found someone to spend his life with
- Good things about the groom
- Something good about the bride (limit this; don't steal the maid of honor's thunder)
- Anecdote or story
- Heartfelt toast (pull one out of any toasts / best man / wedding book at the bookstore, or be creative and make your own)
Can be any length as long as it's from the heart.
My toast was about 3 minutes for a friend's wedding, and it was enough to make all 200 people at the reception start bawling with joy.

I just reminisced about how life passes us by so fast and how we used to horse around as little kids, then said how glad I was that he found such a great woman to be his wife, and also mentioned how nice and hospitable her family was.
Of course, the latter part was utter bullsh!t because her family was a bunch of alcoholic, dysfunctional freaks, and the happy couple is now divorced, but that's beside the point. Live for the moment.
