Want to See Something Microscopic in 3D?

Over the next three week's I'll have access to my school's very expensive (and recently bought) confocal microscope. This is part of a lab module and the Professor is encouraging us to bring in samples. I have some ideas about scoping up some algae or finding some diatoms. Any one of you guys like to request to see something? Samples will look best if they have some kind of auto-fluorescing property (I.E. Green Fluorescent Protein or Chlorophyl). A 3D image is rendered by taking multiple 2D slices of a sample.

Order some ADXL samples from analog devices. Use a soldering iron to take off the metal top. Don't jar the thing, it might break. Take a picture of that - most of it should be a MEMS capacitor. You may be able to see one of the leaves move.

take pictures of a CPU die. That'd be cool to see in 3d due to the vertical layer of copper connections in all of the transistors.

OP, what magification range are you going to be using (10X, 100X, 1000X)?

actually, had you gone ahead and posted the pic in the OP with something like--"Look at this kick-ass confocal image I took--guess what it is!" you would have people creaming all over this thread.

Instead, it's just opened up for some good-natured trolling. Such is the way of AT. :\


that being said, I'd like to see a Drosophila's head. Or, even better, a cross section of 1st instar larvae (Dros.) testis. This is pertinent to my current work. :D

we're trying to section--so, 3-4 cuts--individual testis in fly larvae, and adults (which can be done manually, though it's painful). we want to do gene expression profiles of y chromosome in various developmental stages (each section of testis contains spermatazoa at different developmental stages).

the plan is to get an uber expensive laser microdissection scope to help us with this. Currently, i'm curious about size, magnification needs, general issues of "What do we need to do this?"

Hmm, I know one of the Professors in the Dept. works with Drosophilae. I'll see about getting a head and mounting it. I'll definitely be cool to see the eyes.
