Perhaps some of you can help steer towards some resources regarding this matter:

I'm trying to get high quality video captures for motion analysis. This involves a magnification of about 40-80x, and capture the movement of the object as it bends towards the camera. The problem is that since the magnification is so high, the smallest movement (~100um) would move the reference points out of focus. If anyone can shed some light, that would be greatly appreciated. The magnification is accomplished by attaching the camera into the viewport of a microscope. It is imperative that the capture is done as the rod-like object in question is bend in the middle toward the camera.
