Upgrading RAM in a new iMac

i just sold my MBP and replaced with with a Sandy Bridge iMac. I had a 8GB (2x4GB) RAM upgrade that I was using in my MBP and decided to put it in my iMac. I did and everything is running just fine.

But when I tried putting the RAM that came with my iMac (2x2GB) into the machine with the 2x4GB it just beeped at me. Does it matter which slots I put the RAM in? Can I not mix sizes?
 
You should be able to mix sizes. There are four slots, and I will number them 1 through 4 where one is the furthest left and 1 is the furthest right.

How are the 2*4GB sticks currently arranged
How were the 2*2GB sticks arranged
Are you sure you had the new stick fully inserted

I guess the front and back matters. My 2x 2GB came in the back two slots, and i moved them to the front, and then put 2x 4GB in the back.
