unplug sata with power on?

IF the controller and drive support hot-swapping, yes. I don't think most basic consumer-level ones do (although RAID-capable controllers should if they do RAID1 or RAID5).
 
I would just wait for SATA II. Current SATA hot-swappability is spotty at best right now. Not only does your drive and controller need to support hot-swapping, so does your power supply. If you are using a SATA drive with a standard 4 pin molex plug, you will have problems.
