Why wouldn't it work?

And eSATA is exactly the same as internal SATA, just that it has a different connector.

You can get cables that change eSATA into SATA for using internal hard drives externally.

You can add a whole bunch and you'll be fine. Bear in mind if you're installing an OS on a drive connected to a SATA port on a peripheral card you may need drivers at install time, not sure.
