Troubleshoot a serial port?

I'm having the hardest time trying to get a weather station recognized on the serial port of my pc with XP. The weather station program can't find the serial port and I don't know what to do to make it visible.
I've checked the BIOS to be sure the serial port is enabled, and checked the Device Manager and saw no conflicts.
I disconnected the UPS and disabled the service to be sure it wasn't interfering.

The station came with a loopback connector, and when I plug that in, the station software claims that some other program has COM 1 open (out of the 18 ports it looks at when searching), and can't find the loopback connector in the other ports. I've no idea how to find out what program is using COM 1.

Is there some way to test if the serial port is bad? The MB is an Asus A8V Deluxe

What's the difference between a Serial port & a COM port?
