I'm building my first rig and I wanted to know if I should use as many standoffs as there are holes in the motherboard. If the mobo has 10 holes, should I use 10 standoffs or is that not neccessary?

It's best to use the standoffs the motherboard dictates....esp around ram and the cpu. These keep the motherboard from being cracked or shorting.

Why is this even a question, are you trying to use a case for the wrong type of motherboard?
 
if you have more than needed theres a chance you'll hit one of the things sticking out of the bottom of the mobo and a 99.9% chance it will short as a result
