There's no way for software to tell. As far as the video card is concerned, every pixel was working fine when it shuttled it off to the monitor. It's like a blown tire in a car: the engine will still run just as fast. It doesn't realize it's not going anywhere.
I found the easiest way to detect screwed up pixels is to put up a completely black screen. That'll let you see any stuck in white, green, red or blue. Then, put up a completely white screen, which'll let you see completely dead pixels. This is how I discovered a lone dead pixel (stuck white) on one of my laptops.
And just a note: at 1600x1200 it starts to get really difficult to tell if pixels are dead. My laptop was that resolution, and I didn't notice the pixel for about a month after owning it. It was so small that, in general, I barely ever noticed it.
