There's a wikipedia entry on it, actually. But to sum up, it's just title is just incredibly cool in it's lameness, as is the plot - trying to assassinate someone on a plane by dumping a lot of snakes in the cargo hold. Plus, it's a title and a concept all in one! Really, that's why it's popular. The studios were actually surprised by this internet popularity, but it led them to go back and shoot extra scenes to kick it up from PG-13 to a hard R. They've kicked in the advertising to help push it, since apparently the title itself is helping the movie to get a lot of pre-release interest on the internet, they're probably hoping people not on the internet will also get into seeing the movie just because of the name. They were originally going to change the title to Flight 1234 (or something), but people were pretty vocal about it, and they changed it back. Fans made up the "mvther fvcking snakes, on a mvtherfucking plane thing in fake trailers, because that's the kind of stuff Samuel L. Jackson says in all of his movies. When they went to shoot additional scenes, they made sure he uttered that line.
