PCI express it the new standard for Graphics cards ports, it repleaces the AGP (Accelerating Graphics Port), so it's basically the slot where you plug your card in your motherboard.
SLI (Scalable Link Interface) is the technology of running two graphics cards in parallel using a motherboard that has 2 PCI express 16x ports, which results in 75-100% performance increase over using a single card, and this technology is only available for PCI express video cards, there's no SLI AGP, and the known cards that are SLI capable are 6600GT, 6800GT and 6800 Ultra (all PCIe)
