Resume writing after first job

Just a really quick background: i've been working at my current company, which I joined straight out of college for about 5 years now. The pay's good, the people are good and I have no serious complaints about it (just a useless aside). However, a new really interesting opportunity has come up for international work (same field) which I want to explore.

The point is, I have to update my resume. Given that my original resume is 5 years old and was made prior to any professional experience, i'm more than likely going to have to write a new one from scratch (and bring some much needed formatting updates). I have a ton of great bullet points from my current job (i moved up 3 ranks in four years and accomplished a lot). My questions are as follows:
Does anyone have any experience/advice about creating a resume after the first "professional" job?
Is it odd to only include only one job on the resume? Should I include my previous positions just to have more than one job listing on there or is it generally detrimental if they're unrelated to the field?

As a note on the previous jobs, they were college-level positions (order transcriber/secretary at a hospital) which I picked up only for the money and which have absolutely nothing to do with my current profession.

Any opinions? My primary goal is to make my resume as professional and as impressive as possible to the prospective employers. Normally i'd google this info but, for the life of me, I can't find this scenario.

ditch the stuff you did for part time jobs, it's pretty much worthless unless it shows you working with kids or what not or anything 'out of the ordinary'. But it's just the stuff you listed ignore it.

I would list your 3 roles at your current company as separate jobs so you can separate out the responsibilities. Start and end dates also.

Get in to a habit of updating your cv as and when you add new skills or have new responsibilities.

Thanks for the suggestions. Splitting up my job by titles is definitely a great idea. Good way to organize my achievements.
