I have XP Home on my computer now running on an ATA 133 drive. There is also a 400gb drive with 3 partitions: one for pictures/general documents backup, one for PVR Movie Files, and one for Program files (games and such mostly).

I have a spare SATA drive lying around that has a bit more capacity 100gb vs the 80gb XP is on now, so I want to install XP Home on the spare drive and shelve my 80GB drive. I'm wondering if all the program files from the second hard drive (the 400gb drive currently installed in the machine) will still operate properly, or will I have to reinstall all of them?

Clarification: 2 drives in system now (Primary with OS on it and slave with Program Files on it). Want to replace the primary drive. Will the programs on the slave drive still work, or will they need to be reinstalled?

So your C: is 100% of the 80 gb drive? You might be able to painlessly do a full disk copy of it to the 100gb drive and not to have to install any software. Ghost, PQMagic or Acronis True Image will do the trick. You'll need to make sure that your SATA (100gb) drive is the primary boot drive.
THere's also a free bootabe linux based partition editor/copier that might be able to handle the copy/resize of the ntfs volume. I can't remember what it's called.
 
To be honest I'd rather just start from scratch. The more I think about it the more I'm starting to believe that the Program Files partition should work just the same as the movies partition. The files will still be there, I'll just have to add shortcuts to the Desktop to run them. Either way, I'm not wiping my existing primary HDD, so it's not as if I have anything to lose by just trying this out. I guess that's what I'll do tonight.
 
Those programs that place dll files in the system32 folder or add registry keys at the installation time will not run. An example is Microsoft Office. Another example is Far Cry. Another example is F.E.A.R.

But, there are also programs that will work.
Of course, you need to place a shortcut as you stated.

Your best bet is to create an image of your C and restore it on the new drive.
Or, be prepared to re-install many of your programs.
