Quick memory question

I'm looking to buy another stick of memory for my main pc. I "borrowed" a stick to use while building an HTPC for the den. This results in me having 3 of 4 slots used, so no dual channel at the moment. My question is will I really see that much of a difference to get matching sets of 512mb memory for my main rig (pull ram chip # 3 and put it in the wife's machine) versus just getting another 512 chip to fill the fourth bank? Keep in mind #s 3 and 4 weren't matched to begin with.

Also, if it makes that much of a difference, I'm not a big major overclocker.
