no, four swords is a multiplayer game only..

yes, it will work in singleplayer mode with a regular controller, however, more than 1 player requires a gba for each person

no, 2+ players requires a GBA for every player

yes, you can most certainly play single player with a regular controller, if you do so, the tv screen will pop up a virtual GBA screen for parts where you would normally be on the GBA
