Question about fresh install...

I have an SSD C: drive and an regular E: hard drive...

I need to do a fresh install of windows...

Do I need to format the E: drive (ransomware virus) and then reinstall windows to C: ?

Thanks for any advice...

Seems like we are missing a bit of information, but is e: the old c: drive that got a virus?

If you want to do a fresh install then yes you would need to format c:
If you have a virus that you suspect is on the other drive(old c:) as well I would format that one as well

One the drives are formatted, i would on have the SSD plugged in for the install. This way windows will keep everything on the one drive, where sometimes if it sees a 2nd drive it will put the boot info on the 2nd drive for some dumb reason

C: is my ssd boot drive
d: is my dvd drive
E: is another hard drive

I'll parrot what the others said...

Wipe both of the drives with some sort of tool that will destroy all the data, not just 'format' it, and when clean installing only install the SSD until the OS is up and running.

I hope you had your data backed up... :(
