I have a Diamond mx300 sound card. Last night, I tried my microphone and it worked fine, when I tried recording in sound recorder
Today, I moved the card to another pci slot because I needed my promise ata100 controller closer to my hard drives
The sound cards works fine but now the microphone won't work

I've checked my sound settings, and my microphone is all the way up and the mute is checked

I can't figure out what is wrong.... can anyone help me?

These may sound like really dumb suggestions, but...

<< the mute is checked >>

I hope you mean unchecked :)
Also when you moved the card, you might've put the microphone back into the wrong plug.
Is everything working according to device manager?

no, the mute should be checked
just to be sure, i unchecked it and it still doesn't work
it's plugged into the correct port for the microphone
the sound card works fine - no conflicts in device manager and i can play music and such

yes keep it muted if you do not want to hear your self but unmute to check if its working because you could have your record settings set to record from another source! If all else fails try uninstalling the drivers are reinstalling them!
