Please Educate Me On PCI-E

I am building my first new system in several years and am trying to pick out a video card. I am a Leadtek guy at heart but I am confused about the difference between VGA and PCI-E. I have searched the forums but cannot find anything to set me straight. Can anyone help me out?? Surely there is a website out there you can link me up with.
 
PCI-E is just how the video card connects to the mobo. The counterpart is AGP, which it is replacing. All you need to worry about is getting a mobo that has PCI-E instead of AGP, then getting a video card that is PCI-E, not AGP. If you need a high-end card that is. If not, AGP is still fine, and most cards are still AGP.

I am building a system using an ASUS P4C800-E Deluxe mobo. I don't believe it supports PCI-E but need to know for sure. I am not a heavy gamer but like to have systems that can support more than I do with them. My philosophy is that it is better to have it and not need than need it and not have it. Knowing this, can you recommend a "middle of the road" Leadtek card for the mobo I am using??
 
That is an AGP MB, a good middle of the road card in AGP if you insist on Leadtek is the 6600GT AGP model, its listed at Newegg for $216 but out of stock currently.
