Person A has the ability to visit a certain site at work.
Person B has the ability to visit the same certain site from home.

Little while, person A loses the ability(web site not found) to visit the site at work.
Person B also loses the ability(Website not found) to visit the same certain site from home.
Person B can visit the site from School, Therefore the site is still operative.

What could cause this problem? Any suggestions of ideas, sorry for the lack of Information.

PS: Person B hasn't changed anything to his connection. Person A works at a small company.

Tell A B C D E & F 😛 to all make sure that they are resolving the website to the same IP address. They should be resolving the same IP, if not then you have a DNS issue, possibly cached somewhere (locally or proxy). Now if they are all resolving properly it could be a firewall setting or website being blocked or even an mtu setting on the router.
